Resistance to antibiotic treatments coupled with the decline in discovery has resulted a steady increase deaths caused by once “curable” bacterial infections. Developing new drugs is crucial prevent more loss of life future. We discovered compound resazurin exhibits antimicrobial activity against gram-negative bacteria including Francisella tularensis (Ft), however, certain strains Ft have developed resistance resazurin. Understanding how Ftdevelops will help defining mechanism which elicits its effect. Whole genome sequencing resazurin-resistant (Rzr) LVS mutants revealed 93% isolates sequenced possessed mutations coding regions FTL_0421, FTL_0895, and FTL_1504. The focus my project was explore role FTL_0895 susceptibility. To confirm this gene plays reduced susceptibility Rzr resazurin, wild-type copy cloned into shuttle vector pABST thenelectroporated one mutants, Rzr1.
